DNA sequence reactions done by KIGene 


In order to optimise the sequencing reaction we need the following information:  
  • template and primer concentrations

  • plasmid DNA or PCR fragment?

  • size of the template

  • primer sequence

Material needed:

Purified PCR fragments          

  •  5-50ng depending on length (~2 ng/100bp) in a volume of 5µl or less

Plasmid DNA                   

  • 0.2µg-1.0µg depending on size (~0.2µg/5kb) in a volume of 10µl or less

Primer                                   

  • 5pmol/reaction in a volume of 2µl or less

Sequencing of large clones (BAC, PAC and lambda) per reaction

  • Plasmid DNA - 1.0µg
  • Primer - 30pmol
  • The total volume of plasmid and primer can not exceed 12µl.

When sending the material by post, use padded envelopes and wrap the tubes in Kleenex or similar to ensure they will not be damaged during the postal handling. Please use Parafilm over the lids.
